Progress in this field is summarized. The intracellular localizations, illustrated by electron micrographs, include "Plasma Membrane": nucleoside phosphatases, alkaline phosphatase; Endoplasmic Reticulum": nucleoside diphosphatase, thiamine pyrophosphatase, and, in fewer instances, glucose-6-phosphatase, acid phosphatase and nucleoside phosphatase; "Golgi saccules": nucleoside diphosphatase, thiamine pyrophosphatase, and, in a few instances, alkaline and acid phosphatases; "Lysosomes": acid phosphatase (also esterase); "Mitochondria": "ATPase". Varieties of lysosomes are illustrated. Glutaraldehyde, recently introduced by Sabatini et al., and formaldehyde-calcium are compared as fixatives for electron microscopy of phosphatase activities.
