Neurotensin is a tridecapeptide originally isolated from bovine hypothalamus' and subsequently found to have a widespread distribution within the central nervous system (CNS) and in selected areas of the gastrointestinal tract. Among the earliest reports of the effects of neurotensin on endocrine function were its actions on the anterior pituitary2a3 and the pancreatic islets!.'In our initial studies on the endocrine pancreas, it was apparent that the route of administration was of critical importance since cerebroventricular, in contrast to systemic, injection of the peptide did not affect levels of plasma glucagon, insulin, or glucose. 5 We therefore initiated a series of studies to determine whether the pituitary hormone-releasing effects of neurotensin involved central or peripheral mechanisms and to explore the possible mediating role of other centrally active compounds. This report summarizes the experience from our laboratory related to the effects of neurotensin on growth hormone (GH), thyroid-stimulating hormone (TSH), and prolactin (PRL) release and on the mechanism of its action.
