Purpose: The present study aimed to observe the effect of hippocampal low-frequency stimulation (Hip-LFS) on the GABAA receptor α1, α5 and γ2 subunit expression in amygdalakindled pharmacoresistant epileptic rats. Methods: Twelve pharmacoresistant epileptic rats were selected from 65 of a kindled model.of epilepsy, and were assigned to a pharmacoresistant control group (PRC group, 6 rats) or a pharmacoresistant stimulation group (PRS group, 6 rats). The pharmacosensitive epileptic rats were used as a pharmacosensitive control group (PSC group, 6 rats). A stimulation electrode was implanted into the hippocampus in all the groups. The Hip-LFS was administered twice per day for 2 weeks. Then, the GABAAR α1, α5 and γ2 subunit expression in hippocampal tissues was measured by a real-time quantitative polymerase chain reaction (PCR) and Western blot. The seizure degree and the after discharges were also measured. Results: The stimulation-induced seizures and the after discharges were inhibited significantly after Hip-LFS. The GABAAR α1, α5 and γ2 subunits in the PRC group decreased dramatically compared with that in the PSC groups. The GABAAR α1, α5 and γ2 subunits (both mRNA and protein) increased significantly compared with the PRC group. Conclusions: The Hip-LFS might be effective in the amygdala-kindled model of pharmacoresistant epilepsy. The antiepileptic effects of the Hip-LFS may be achieved partly by increasing the GABAAR α1, α5 and γ2 subunits.
