MATERIALS AND METHODSAmerican bullfrogs (Rana catesbeiana) were stored at 4OC in saline. After section of the spinal cord, the proximal 2-cm segment of duodenum was excised, and the seromuscular layers were removed. The mucosa was mounted in a Lucite chamber of l-cm2 exposed area and bathed by solutions at room temperature circulated by gas-lift systems. The nutrient (serosal) solution contained (mM): Na+, 102.4; Cl-, 91.4; K’, 4.0; Mg2+, 0.8; Ca2’, 1.8; H2P04, 0.8; SO:-, 0.8; HCO;, 17.8; and glucose, 2.0 and was gassed with 95% 025% COZ. The isosmolar, unbuffered secretory (mucosal) solution contained (mM): Na+, 102.4; Cl-, 91.4; K+, 4.0; Mg2+, 0.8; Ca2’, 1.8; S@, 10.1; and mannitol, 11.3 and was gassed with 100% 02. Cl-free solutions were made by substituting S@-in the secretory solution and isethionate in the nutrient solution. Na+-free solutions were made by substituting choline for Na+. HCOF-free solutions were buffered with 5 mM N-2-hydroxyethylpiperazine-N’-ethanesulfonic acid (HEPES) and gassed with 100% 02. Anoxia was achieved by gassing the nutrient solution with 95% N2-5% Co2 and the secretory solution with 100% N2. The nutrient pH was 7.23. Alkaline secretion was measured in the secretory solution
