Syncytium-forming viruses from bovine, feline, and simian species grew to highest titers in dividing cell cultures from a variety of species and had similar cytopathic effects. Both intranuclear and cytoplasmic antigens were detected by immunofluo-rescence. An 8-day plaque assay was developed, and the growth cycle of simian “foamy” virus type 3 was determined; there was a relatively prolonged latent period of 26 hr. Even with optimal conditions, virus yields were low and did not exceed 10 plaque-forming units/cell per 24 hr. Like other viruses with an RNA-directed DNA polymerase, the formation of infectious simian “foamy” type 3 was markedly inhibited by low concentrations (10 μg/ml) of the thymidine analog, BrdU; similarly “foamy” virus infectivity was also extremely resistant to inactivation by ultraviolet light, suggesting another shared biological feature of viruses with a reverse transcriptase.
