Frozen sections of unembedded, fixed and fresh nerve fibers which are thin enough for electron microscopy can be prepd. with a described modification of the thermal-expansion technique of Newman, Borysko and Swerdlow. Examination of longitudinal and transverse frozen sections of rat sciatic nerve fibers fixed in osmic acid or formalin discloses the following structures The bulk of the myelin sheath breaks down into piles of thin, trasparent and flexible lamellae, approx. 50 A thick, with a characteristic granular surface structure, 50-100 A high. Transverse sections through nerve fibers reveal 2 distinct fibrous layers adhering closely to the central, lamellar portion of the sheath: (a) an external granular membrane covered with typical collagen fibrils and corresponding to the Neurilemma and the adjacent connective tissue. A special type of extremely thin smooth fibrils, 100 A thick and with no detectable periodicity, bound together in compact strands, is regularly seen attached to the collagen fibrils of this outer layer. (b) An internal layer of densely woven thin fibrils (100 A thick) which connects the myelin sheath with the fibrous structures of the axon. The axon contains a fine network of fibrils which are approximately 100 A in width and are stretched out in a transverse or oblique transverse direction.
