C. floridanumfirst instar larvae isolated from laboratory-rearedPseudoplusia includens[Chrysodeixis includens] andTrichoplusia niwere incubated for 30 minutes in phosphate buffer saline (PBS; 7.2 pH) with or without dispase (1 U/100 µl). The larvae in PBS remained envelope by the membrane. After rinsing in PBS, the larvae were inoculated into permissive (P. includensandT. ni) and non-host (Spodoptera frugiperda, Heliothis virescens, andManduca sexta) species. All hosts were dissected 24 h post-inoculation, and the number of recovered larvae that were encapsulated was determined by phase-contrast microscopy. In general, the presence or absence of the syncytial membrane strongly affected the encapsulation rate in all host species. The larvae without an extraembryonic membrane were encapsulated irrespective of host species. The membrane-enveloped larvae were almost never encapsulated inP. includensorT. ni, but an increasing proportion of larvae were encapsulated in the non-host species. The larvae enveloped by a syncytial membrane elicited the weakest encapsulation response inP. includensandT. ni, an intermediate response inH. virescens, and the strongest responses inM. sextaandS. frugiperda. The inspection ofC. floridanumrecovered fromP. includensandT. niindicated that no granular cells bound to the membrane-enveloped larvae, but granular cells and plasmocytes were present in the capsules that formed around the larvae without a membrane. This suggests that permissive hosts may not encapsulateC. floridanumbecause granular cells are unable to bind to this surface. The absence of such protection in non-host species suggests that variation exist in the recognition and/or adhesion receptors on haemocytes that mediate the recognition ofC. floridanum.
