It is now widely believed that the high concentration of K+ ions and the low concentration of Na+ ions found in most animal cells depend on the activity of a pump, in the membrane, which uses energy from the hydrolysis of adenosine triphosphate (ATP) to accumulate K+ ions and expel Na+ ions. The aim of this review is to summarize the evidence that justifies this belief, and to give a brief account of recent work that has provided a little information about the nature of the pump mechanism. For more detailed reviews covering the period up to 1965 see Skou (1965) and Glynn (1966); a review by Heinz (1967) provides a useful source of recent references.
