The potassium salt of 1-hydroxy-7-azabenzotriazole can be used along with FmocNHCH(R)COCl (Fmoc = 9-fluorenylmethoxycarbonyl; R = amino acid side chain) as coupling agents in peptide bond formation. The acylation reactions are rapid and efficient and can be carried out in an org. medium. The yields as well as purity of the peptides are satisfactory. For example, Fmoc-Val-Gly-OCH2Ph was synthesized in 88% yield from Fmoc-Val-Cl and H-Gly-OCH2Ph·HCl.
