A method is descr. for the study of the consistency of the cytoplasm of cells in the outgrowth of tissue cultures. Magnetic particles are phagocytosed by the cells; these particles are rotated within the cell by an external magnetic field, and their displacement is recorded by the cine-microscope. It has been shown that the cytoplasm of chick embryonic osteoblasts is a weakly elastic fluid, with a modulus of rigidity of the order of 102 dynes/cm2. There is evidence of thixotropic behavior. These results do not support theories of protoplasmic structure which postulate an interlacing network of fibrous micelles. It is suggested that the fine structure of the cytoplasm of these cells is much more varied and labile.
