Wolbachia‐induced reproductive regulation in hosts has been used to control pest populations, but little is known about the molecular mechanism underlyingWolbachiaregulation of host genes. Here, reproductive regulation byWolbachiain the spider miteTetranychus truncatuswas studied at the molecular level. Infection withWolbachiaresulted in decreasing oviposition and cytoplasmic incompatibility inT. truncatus.Further RNA‐seq revealed genes regulated byWolbachiainT. truncatus. Real‐time quantitative polymerase chain reaction (qPCR) showed that genes, includingchorion protein S38‐likeandRopwere down‐regulated byWolbachia. RNA interference (RNAi) ofchorion protein S38‐likeandRopinWolbachia‐uninfectedT. truncatusdecreased oviposition, which was consistent withWolbachia‐induced oviposition decrease. Interestingly, suppressingRopinWolbachia‐infectedT. truncatusled to increasedWolbachiatitres in eggs; however, this did not occur after RNAi ofchorion protein S38‐like. This is the first study to show thatchorion protein S38‐likeandRopfacilitateWolbachia‐mediated changes inT. truncatusfertility. In addition, RNAi ofRopturned the body colour ofWolbachia‐uninfectedT. truncatusblack, which indicates that the role ofRopis not limited to the reproductive regulation ofT. truncatus.
