A disulfide-containing analog of bis-acrylamide was synthesized. When substituted for bis-acrylamide as the cross-linking agent in polyacrylamide-gel formation, a gel is formed which has electrophoretic properties that are quite similar to normal gels. Gels formed with the disulfide cross-link are soluble in the presence of 2-mercaptoethanol under conditions which do not degrade RNA. RNA samples [from Bacillus cereus] which had been separated electrophoretically were recovered in about 65% overall yield from the dissolved gel by ion-exchange chromatography. The reduced polymer can be re-gelled by atmospheric oxidation. Disulfide cross-links provide versatility and control of the physical state of the gel matrix which were not previously available.
