Recovery of the gonadal-pituitary axis in intact adult male rats was investigated for 15 days after a single injection of estradiol benzoate (EB), and for 60 days after 14 daily injections of EB. After a single injection of EB, plasma and testicular levels of testosterone (T) were suppressed for 2 days, but recovered to control levels by Day 4 and thereafter. Plasma LH, however, was significantly suppressed 4 and 6 days into recovery, while T had returned to normal values. During this time, the mean pituitary content and concentration of LH were decreasing and, although these decreases were not statistically significant, they may point to possible EB suppression of LH synthesis. On the other hand, plasma FSH significantly decreased 2 days after treatment, while pituitary FSH concentration and content were significantly increased, indicating that EB suppressed the pituitary release, but not the synthesis of FSH.The recovery patterns of T and gonadotropins seem to indicate that EB may act independently on the testis and the pituitary since the testis recover to control levels more rapidly than does the pituitary. The data also indicate that EB acts on the pituitary of intact rats by 2 different mechanisms; one affecting FSH release and the other affecting LH synthesis. The single dose of EB did not produce any changes in spermatogenic activity or Leydig cell morphology of the testes.After chronic EB treatment, the suppressed plasma and testicular concentrations of T did not recover to normal levels throughout the 60-day period. Pituitary LH was significantly suppressed for 2 weeks after treatment, with no drop in plasma LH. A rebound in both pituitary and plasma LH occurred by 3–4 weeks after termination of EB treatment. The increases in LH were not accompanied by elevations in T. The initial suppression of plasma and pituitary FSH at 2 and 6 days after cessation of EB treatment was followed by a rebound similar to that of LH. The spermatogenic activity in the testes was affected only the first 10 days after chronic treatment with EB, but recovered normally thereafter.
