The molecular components of cytoplasmic microtubules analyzed in situ in several types of cultured cells [baby hamster kidney BHK cell, hamster fibroblast NIL cell, hamster fibroblast NIL-HSV cell, mouse fibroblast 3T3 cell, mouse neuroblastoma NB 2A cell, rat glial tumor C6 cell, human cervical cancer HeLa cell, human MF 7 cell and chicken embryo fibroblast CEF cells.] This approach made use of preparations of detergent-extracted cytoskeletons that contain the original cellular microtubules. Identification of microtubule components is based on 2 criteria; their presence in the cytoskeleton prepations is dependent upon the presence of intact microtubules and they are released from such preparations by buffers that depolymerize microtubules. Proteins that meet these criteria are described. Some are restricted to cells of certain species or tissues of origin. Others appear to be of widespread distribution. Among the latter is a protein homologous to one of the [calf brain] tau polypeptides in brain microtubule protein.
