The pharmacological classification of dopamine receptors is controversial. While as many as five subclasses of dopamine receptors have been proposed, in this review we attempt a synthesis which suggests that the preponderance of data can be encompassed by a model proposing only two major classes of dopamine receptors. Hopefully, this review will provide the reader with a useful conceptual framework within which to evaluate future research on dopaminergic pharmacology.Considerable experimental evidence, both biochemical and pharmacological, indicates that multiple classes of dopamine receptors are present in vertebrate and invertebrate nervous and endocrine systems. Classifications of these receptors may be based on behavioral, physiological, anatomical, biochemical and/or biophysical criteria. However, each method of classification must also integrate pharmacological criteria; that is, how diffenmt drugs interact with each discriminating response or putative structural subtype of receptor mediating that response. Of course, the ultimate goal of pharmacology is to integrate knowledge about the action of agents at all the levels of inquiry mentioned above.
