Proteomics identification of macrophage-derived signals impacting metaflammation in tissues (P11)

巨噬细胞(MΦ)来源的信号调节组织的动态平衡，并在代谢过程中驱动病理生理学。然而，由于以往在体内全面研究细胞间信号的技术限制，这些信号很少被发现并从机制上进行研究。本项目旨在发展基于质谱学的蛋白质组学方法，包括细胞类型特异性标记，以剖析MΦS在体内高脂饮食过程中组织驻留细胞和浸润性细胞之间的通讯。我们将对组织MΦ信号生成一个系统水平的视图，重点放在肝脏上，并为针对MS以减弱或逆转代谢反应的新假说提供基础。

Macrophage (MΦ)-derived signals regulate tissue homeostasis and drive pathophysiology in meta-flammation. Due to previous technical limitations in studying the intercellular signals comprehensively in vivo, however, few of these signals have been discovered and studied mechanistically. This projects aims to develop mass spectrometry-based proteomics methods, including cell-type specific labelling, to dissect the communication between MΦs, tissue-resident and infiltrating cells during high fat diet in vivo. We will generate a system-level view on tissue MΦ signals with a focus on the liver and provide the basis for new hypotheses to target Ms in order to attenuate or revert metaflammation.
