Mechanism and regulation of gastric carcinogenesis directed by the Helicobacter pylori CagA oncoprotein

幽门螺杆菌CagA阳性菌株在胃癌的发生发展中起关键作用。本项目阐明了1)幽门螺杆菌将其癌蛋白CagA送入宿主胃上皮细胞的机制：2)鉴定了一种人类蛋白Pragmin，该蛋白的功能与所传递的CagA：相似；3)在慢性感染cagA阳性的幽门螺杆菌时，已发现一条分子通路，可将胃上皮细胞重新编程为类肠道细胞。最后，对幽门螺杆菌CagA蛋白进行了结晶，并通过X-射线分析确定了其三维结构。

Helicobacter pylori CagA-positive strains play a key role in thedevelopment of gastric carcinoma. This project elucidated 1) the mechanism by which H. pylori delivers its oncoprotein CagA into host gastric epithelial cells: 2) identified Pragmin,a human protein the function of which is mimicked by delivered CagA: and 3) uncoveredmolecular pathway that directs reprograming of gastric epithelial cells into intestinal-likecells upon chronic infection with cagA-positive H. pylori. Finally, the H. pylori CagAprotein was crystalized and its three-dimensional structure was solved by X-ray analysis.
