Role of Epidermal Grpwth Factor in Human Endometrial Decidualization.

结果如下：(1)对人蜕膜组织中表皮生长因子(EGF)进行了免疫组织化学定位观察。(2)免疫沉淀-放射自显影显示蜕膜细胞产生EGF。(3)Scatchard图显示蜕膜细胞只有一种EGF受体，其Kd值为1.02 nM，Bmax为89.4 pm/2×10~(-5)细胞。(4)EGF剂量依赖性地抑制蜕膜细胞催乳素的生成，IC_(50)为700pg/ml。(5)EGF对催乳素的抑制作用是由酪氨酸激酶和蛋白激酶C诱导的。(6)EGF通过激活蛋白激酶C而促进特异性蛋白磷酸化。

The results were as follows ; (1) Immunohistochemical localization of epidermal growth factor (EGF) were observed in decidualized human endometrium. (2) Production of EGF by decidual cells was clearly demonstrated immunoprecipitation-autoradiography. (3) Scatchard plots revealed decidual cell possesses single kind of EGF receptor with Kd of 1.02 nM and Bmax of 89.4 pM/2x10^5 cells. (4) EGF dosedependently inhibited prolactin production by decidual cells with IC_<50> of 700 pg/ml. (5) The inhibitory effect of EGF on prolactin production was induced by the tyrosine kinase and protein kinase C. (6) EGF enhanced specific protein phoshorylation (48kDa, acidic protein) by the activation of protein kinase C.
