Basic and clinical analysis of basic FGF in the regenerating liver

本项目旨在研究再生大鼠肝脏中bFGF结合位点的定位及其抑制因子，以及肝切除术过程中炎症和抗炎细胞因子的变化。免疫印迹实验显示再生肝核亚细胞部分中bFGF受体瞬间升高，125 I-bFGF结合实验显示再生肝核中bFGF特异性结合位点和结合能力较对照大鼠增加。术中炎性细胞因子如il - 6、TNF - α及抗炎细胞因子如il - 10均升高。肝切除术期间血清bFGF未见明显变化。这些结果提示bFGF和炎症因子在肝细胞增殖和有丝分裂中起重要作用。

This project was performed to investigate the localization and its inhibitory factor of bFGF binding sites in regenerating rat liver, and the changes of inflammatory and antiinflammatory cytokines during hepatectomy. Immunoblot experiments showed a transient increase of bFGF receptor in the nuclear subcellular fraction of regenerating liver and 125 I-bFGF binding assay indicated specific bFGF binding sites in nuclei and binding capacity increased in regenerating liver than that of control rats.Both Inflammatory cytokines such as IL6, TNF α and antiinflammatory cytokine such as IL10 were increased during operation. No definite changes of serum bFGF was detected during hepatectomy.These results suggest an important role of bFGF and inflammatory cytokines in hepatocyte proliferation and mitosis.
