Restoration of self liver using transplantation of autologous cryopreserved hepatocytes.

今年，我们从转移性肝癌患者的切除肝脏标本中分离出人肝细胞。经过对分离方法的改进，我们可以获得完整的人肝细胞用于移植。我们将这些肝细胞移植到裸鼠和SCID小鼠体内，并采用我们原有的方法，通过对宿主肝脏进行辐射来增殖移植的肝细胞。利用这些技术，移植的人肝细胞在生长优势后，增殖非常好，直到16周时替代宿主肝脏达到95%。在移植的人肝细胞周围，观察到间隙连接和糖原生成。虽然自体肝移植尚未实施，但肝细胞储备和再次移植的概念可能是终末期肝病的一个很好的治疗选择。

This year we isolated human hepatocytes from resected liver specimen from patients with metastatic liver cancer. After some modification of the method of isolation, we could obtain intact human hepatocytes for transplantation. We transplanted those hepatocytes in nude mice and SCID mice.In addition, we used our original method to proliferate transplanted hepatocyte using radiation on host liver. Using those technique, after growth advantage, transplated human hepatocytes proliferate very well until replace a host liver to 95% By the time of 16 weeks. Around transplanted human hepatocytes, gap junction and glycogenesis were observed. Although auto transplantation has not been performed, concept of hepatocyte banking and retransplantation could be a good therapeutic choice for end stage liver diseases.
