Bei einer wachsenden Zahl Mikrotubuli-assoziierter Proteine konnte eine spezifische Konzentrierung an Mikrotubuli-plus-Enden gezeigt werden. Dort bilden sie einen hochdynamischen supramolekularen Komplex einer Größe von mehr als 3 MDa aus. Der Mikrotubuli-plus-End-Komplex wird vermutlich für das Verankern der Mikrotubulispitzen an cortikalen Bindungsstellen benötigt, wobei er Wechselwirkungen sowohl mit cortikalem Aktin als auch mit Membranproteinen vermittelt. Dieser Vorgang spielt eine wesentliche Rolle bei der Zellkernmigration, der Spindelorientierung und gerichteter Zellmotilität. Wir verwenden Dictyostelium Amöben als Modell, da diese sich durch schnelle Zellmigration und gute Zugänglichkeit für genetische Manipulationen auszeichnen. Mit DdEB1, DdCP224 und der Dynein-Intermediärkette haben wir bereits drei Marker des Mikrotubuli-plus-End-Komplexes charakterisiert. Unsere Ergebnisse deuten darauf hin, daß solche cortikalen Wechselwirkungen die Wanderungsrichtung und die Positionierung des Mikrotubuli-Organisationszentrums (MTOC) in amöboiden Zellen bestimmen könnten. Unser Projekt soll die Rolle von DdLIS1 und DdGlued bei diesem Vorgang klären. Unsere Ziele innerhalb der ersten Zweijahresperiode des SPP1128 sind eine eingehende subzelluläre Lokalisation beider Proteine und die Analyse ihres dynamischen Verhaltens in beweglichen lebenden Zellen, unter Anwendung der fortschrittlichsten Methoden der Lichtmikroskopie. Parallel, planen wir eine funktionelle Charakterisierung von DdLIS1 und DdGlued durch die Untersuchung entsprechender Deletions-, Null- und Über/Unterexpressionsmutanten. Summary: A growing number of microtubule-associated proteins have been shown to accumulate specifically at the plus ends of microtubules where they form a highly dynamic supramolecular complex with a size of more than 3 MDa. The microtubule plus end complex seems to be required for the capture of microtubule tips at cortical sites by mediating interactions of microtubule tips with cortical actin as well as with membrane proteins. This process plays a major role in nuclear migration, spindle orientation and directional cell movements. We use Dictyostelium amoebae as a model since they exhibit fast chemotactic cell migration and are easily susceptible to genetic manipulation. With DdEB1, DdCP224 and the dynein intermediate chain, we have already characterised three markers for the microtubule plus end complex. Here we intend to focus on DdGlued and DdLIS1, the Dictyostelium homologues of p150glued (= dynactin 1) and the Miller-Dieker Lissencephaly protein LIS1, respectively. In mammalian cells, both are found at microtubule plus ends and seem to play an important role in the interaction of microtubule tips with the cell cortex. Our data suggest that such cortical interactions could determine the direction of migration and the positioning of the microtubule-organizing centre (MTOC) in migrating cells. Our project should shed light on the role of DdLIS1 and DdGlued in this issue. Our goals within the first 2-year period of the SPP1128 are a detailed subcellular localisation of the two proteins and live analysis of their dynamic behaviour in motile cells using most advanced light microscopic techniques. In parallel, we aim at a functional characterisation of DdLIS1 and DdGlued by analysis of deletion, knockout and over/underexpression mutants. Furthermore, we intend to examine cytosolic complexes containing DdLIS1 and DdGlued at the biochemical level.
