Inhibition of biofilm formation focused on de novo biosynthesis of purine nucleotides

研究了嘌呤核苷酸从头合成对荧光假单胞菌Pf0-1生物膜形成的影响。对基因敲除菌株的分析表明，从磷酸核糖-1-焦磷酸到5‘-单磷酸肌苷途径中的8个基因的中断导致生物被膜的形成显著减少。对突变菌株的菌落形成单位数的测定表明，大多数活细胞存在于生物膜中。抑制这一生物合成途径不利于细菌在培养基中的生长，但突变菌株仍能形成生物膜。

Role of de novo biosynthesis of purine nucleotides on biofilm formation by Pseudomonas fluorescens Pf0-1 was investigated. Analysis of knockout strains revealed that disruption of eight genes in the pathway from phosphoribosyl-1-pyrophosphate to inosine 5’-monophosphate resulted in significant decrease in biofilm formation. Determination of numbers of colony forming units for the mutant strains indicated that the majority of viable cells were found in biofilm. Inhibition of this biosynthetic pathway was detrimental to bacterial growth in the medium, but the mutant strains were still able to form biofilm.
