The CSIRO Australian National Fish Collection (ANFC) specialises in temperate marine fishes from the Australasian region including those from the Australian Exclusive Economic Zone. Specimens and samples are available to the community for diverse research purposes, however like most fluid-filled collections, the ANFC specimens have been fixed primarily in 10% formalin and preserved in ethanol. Mini-barcoding assays of mitochondrial DNA (mtDNA) cytochrome oxidase subunit 1 (COI) and 16S ribosomal RNA (16S) genes were undertaken with Sanger sequencing and two-step tagged PCR amplicon sequencing on an Illumina MiSeq with good quality and degraded DNA (from formalin preserved tissues) for species identification from our reference collection.
