This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This protocol describes and supports our recently funded American Cancer Society Targeted Research Directed at Poor and Underserved Populations Grant entitled: "hMSH2 and hMLH1 Mediated Colorectal Cancer Among African-Americans" a study designed to analyze the contribution of the DNA mismatch repair genes to colorectal cancer incidence among African American. This Protocol also describes a direct and logical extension of the research described for the African-American population in our ACS funded project. This extension is designed to include all individuals and families at increased risk of developing colorectal cancer regardless of national or ethnic origin. The mismatch repair gene literature to date has predominantly focused on highly selected, extensive pedigrees characterized by multiple affected individuals. A population based analysis of mismatch repair gene associated colorectal and other related malignancies remains unreported. This Protocol will facilitate the identification and molecular genetic characterization of families and individuals at increased risk of colorectal cancer without restriction to highly selected pedigrees; a novel contribution. This effort will clarify the contribution of the mismatch repair genes to overall colorectal cancer incidence. It will also facilitate future efforts to identify novel genetic factors associated with colorectal cancer predisposition as well as factors that influence response to chemoprevention, primary treatment and natural history.
