This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The Fluorescence Imaging Core provides researchers with analyzable images of cells and tissues. Equipment includes a Nikon E-800 fluorescent microscope, high resolution video and a computer- driven image capture and analysis system. Additionally, a BioRad Radiance 2000 confocal microscope and imaging system can be utilized for determining the subcellular localization and distribution of specific proteins in tissue slices or cultured cells. NIH Imaging, Image J, Image Pro Plus v6, LaserSharp and Photoshop software is available. We offer routine fixation, embedding, sectioning and staining of tissue specimens. The laboratory is fully equipped to prepare paraffin or frozen embedded tissue for enzyme histochemistry, immunohistochemistry and immunofluorescence. Our tissue processing and histology procedures are designed to optimize tissue fixation, optimize preservation of antigens and cellular detail and orient tissue for sectioning for the best structural analysis. The Histology laboratory maintains a Shandon Finesse 385 and RMC MT-920 microtomes for cutting paraffin embedded tissue, a Shandon Citadel 2000 tissue processor, a Shandon Histocentre embedding station, a Shandon Veristain 24-4 automated stainer and Shandon Cryotome microtomes for frozen tissues( one each for human and non human tissue). The lab also has available slides, coverslips, mounting media, waterbaths and a variety of other histology tools.
