Heavy metal is one of the main causes of human reproductive diseases and animal reproductive disorders. Nevertheless, the relevant reproductive toxicological mechanisms are not yet clear. The applicant's previous researches demonstrated that cadmium ions can inhibit acetylation of proteins in mouse testicular spermatogenesis, and affect the replacement processes of Tnps and Prms by histones, which suggested that protein acetylation may be involved in cadmium-mediated inhibitions of mouse spermatogenesis and sperm maturation. To elucidate the molecular mechanism of spermatogenesis damage mediated by cadmium-regulated protein acetylation, based on our previous Cd2+ reproductive toxicology and acetylation proteomics research, we utilize cell transcriptomics, metabolomics, modified peptide antibody affinity enrichment technology and ChIP-seq technologies to systematically study the changes of protein acetylation during spermatogenesis, differentially expressed genes and TCA metabolites upon Cd2+ exposure. We will explore the mechanism of Cd2+-induced inhibition of protein acetylation modification during spermatogenesis, and focus on analyzing the association between H4K5 and H4K12 acetylation inhibition and transcriptional expression of related genes, and exploring the interaction between acetylation inhibition by Cd2+and the replacement of Tnps and Prms by histones. This project aim at systematically clarifying the molecular mechanism of sperm cell damage mediated by cadmium-inhibited protein acetylation in mouse. The significance of the project is to provide theoretical basis for solving the scientific problem of Cd2+-reproductive toxicology mechanism, and provide novel sights for the prevention and treatment of toxic diseases by heavy metals.
金属是人类生殖疾病及动物繁殖障碍的关键诱因之一,然而相关生殖毒性的分子机制尚未明确。为阐明Cd2+调控蛋白乙酰化介导生殖细胞损伤的分子机制,本项目在前期研究基础上,利用空间转录组学、代谢组学、修饰肽段富集及Chip-seq技术,系统研究Cd2+暴露下睾丸生殖细胞蛋白乙酰化修饰、差异表达基因及TCA代谢产物变化,探究Cd2+抑制生殖细胞蛋白乙酰化修饰的作用机制。研究结果如下:1)建立Cd2+暴露下生殖细胞损伤动物模型,发现Cd2+处理后ATF3、NRF2和HO-1呈剂量依赖性上调,而KEAP1表达下降,证实在Cd2+暴露条件下触发了NRF2/HO-1信号通路,导致生殖细胞发生铁死亡;2)空间转录组分析结果显示Cd2+处理后无论是各细胞亚群还是睾丸细胞总数显著低于对照组;从整个细胞图谱来看,不同细胞系基因表达明显分成4类基因集群;在Cd2+作用下间质细胞及支持细胞Gbp4、Gbp5、Zbp1及C3基因表达量显著升高,而圆形细胞及精子细胞中Ak1基因表达量显著降低;3)共发现356个蛋白质上的493个乙酰化位点显著上调,271个蛋白质上的503个乙酰化位点显著下调,其中线粒体中酶蛋白乙酰化修饰均显著下调。Cd2+处理后TFRC中K380乙酰化修饰水平显著高于对照组并促进细胞发生铁死亡;4)Cd2+处理组在组蛋白N端23个位点乙酰化修饰水平显著降低,核心区域3个位点乙酰化修饰水平显著上调;5)Cd2+处理后HATs和HDACs转录和蛋白质表达水平无显著变化,但HDAC1 K200位点乙酰化修饰水平显著升高,发现K200乙酰化修饰水平与其酶活性存在显著正相关,干扰PRM2替换组蛋白过程,继而影响小鼠精子发生;6)Cd2+导致H3K18乙酰化修饰水平下降;该修饰位点关联基因转录分析结果证实,H3K18乙酰化修饰可以结合于Pink1的启动子区域发挥转录调控作用,调控生殖细胞线粒体自噬流,进而影响精子细胞的形成。
