基于LPS/TLR4/NF-KB信号通路研究热敏灸大椎穴干预哮喘大鼠气道炎症反应的效应机制

热敏灸治疗哮喘（慢性持续期）较传统悬灸更有疗效优势，但其效应机制研究尚不明确。哮喘发病机制与LPS/TLR4/NF-KB信号通路密切相关。前期研究发现悬灸哮喘大鼠能改善大鼠行为学及肺组织病理学变化，减轻哮喘发作。因此，本研究以哮喘（慢性持续期）大鼠模型为观察对象，以红外成像为热敏穴检测技术，以艾灸“大椎”穴为干预措施，LPS/TRL4/NF-kB信号通路为研究切入点，运用Real time-PCR、Western blot、免疫荧光技术等多种技术手段，从TRL4、MyD88、TRAM、IkB磷酸化、NF-kB等与LPS/TRL4/NF-kB信号通路多个环节的复杂关系中，探讨热敏灸对哮喘（慢性持续期）气道炎性反应治疗作用的效应机制，为热敏灸治疗哮喘（慢性持续期）临床应用提供部分实验依据。

Heat-sensitive moxibustion is more effective in treating bronchial asthma (chronic duration) than traditional suspension moxibustion, but the research on its effect mechanism is still unclear. The pathogenesis of bronchial asthma is closely related to the LPS/TLR4/NF-KB signaling pathway. The previous study found that heat-sensitive moxibustion can reduce asthma attack through improve the behavioristics and lung histopathological changes of rats Therefore, In this research, the model of asthma (chronic duration) rats is used as the observation object, the infrared imaging is used as the detection technology, with the moxibustion "Dazhui"(DU14) acupoint as the intervention measure. LPS/TRL4/NF-kB signaling pathway is the research entry point. It is to explore the possible mechanism of heat-sensitive moxibustion for the treatment of asthma (chronic duration) by using Real time-PCR, Western blot, immunofluorescence technology. It is complex relationships with signaling pathway for TRL4, MyD88, TRAM, IkB phosphorylation, NF-Kb, TNF-α, IL-4, ICAM-1 and iNOS. It is for airway inflammatory response to provide some experimental basis for the clinical application of heat-sensitive moxibustion for asthma (chronic duration).
