环是自然界中重要的结构。聚合物自组装可以制备环状结构,但是通常与棒等其它结构共存。目前尚未掌握聚合物自组装成环的控制规律和内在机理。在前期研究中,申请人发现了棒状胶束弯曲环化制备规整环状结构以及聚合物直接自组装形成螺旋表面环状结构的新现象。基于这些环状组装体制备的研究,本项目拟系统开展环状组装体的制备、表面图案结构调控及其在细胞膜上内嵌成孔行为的研究。采用直接组装和间接组装两种方法制备表面图案化环状组装体。在直接方法中,聚肽嵌段共聚物与聚肽均聚物共混自组装,形成表面图案化环状组装体;在间接方法中,先由聚肽接枝共聚物自组装形成环,然后聚肽嵌段共聚物在环表面自组装形成不同纳米图案。在掌握环状组装体的可控制备以及表面图案调控规律的基础上,开展环状组装体与细胞相互作用的研究,探索环状组装体在细胞膜上成孔的条件和规律。另外,结合实验研究开展理论模拟计算,深化实验观察并指导实验研究。

The toroid is a fundamental geometrical structure in nature. Polymers are capable of assembling into toroidal structures, however, they usually coexisted with rod-like and other morphological structures. Currently, the formation mechanism for the toroids has not been explored. In the previous work of the applicant, toroidal structures have been prepared through a supramolecular cyclization of rod-like micelles (producing toroid exclusively) and self-assembly of a mixture of block copolymers and homopolymers (coexisting with rod-like structures). In this proposal, based on the applicant’s previous work, we aim to systematically study the formation of the toroids and surface patterning of the toroids, and seek the possibility of these surface-patterned toroids as a hole-formation agent on the cell membrane. Two approaches, i.e., direct and indirect approaches will be applied to construct the surface-patterned toroidal nanostructure. In the direct way, the toroids with various surface patterns are self-assembled from polymer mixtures containing polypeptide block copolymers and polypeptide homopolymers. In the indirect way, toroidal nanostructures are first self-assembled from polypeptide graft copolymers through a rod-like micelle-toroid route as reported in our previous work. Subsequently, polypeptide block copolymers self-assemble with the toroids constructing various surface patterns on the toroids. Through these researches, the formation mechanism for the toroidal nanostructures as well as the surface patterns will be revealed, and the method for constructing the surface patterned toroidal nanostructures will be governed. On the basis of the controlled preparation of the surface patterned toroidal nanostructures, we intend to investigate the interactions between the toroids and cells focusing on the hole-forming property of the toroids. By studying, the structure characters of the toroids and the surface patterns facilitating the hole forming on the cell membrane will be summarized. In addition to the experimental studies, theoretical simulations will be carried out to explore both the self-assembly mechanisms and the insertion and hole-forming behavior of the toroidal nanostructures. The results gained from the studies could provide valuable information and guidance for the controllable preparation of surface patterned toroidal structures as well as the further applications.
本项目中,按照计划开展了相关研究,包括聚肽共聚物和均聚物的合成与表征、聚肽均聚物和共聚物的自组装成环行为与机理、聚肽纳米环的细胞内化行为及其机理等研究。在实验研究的基础上,结合理论模拟计算,进一步深化了对自组装机理的探究。整体上,完成了预定的研究目标。.通过聚肽均聚物在有机溶剂/水体系中自组装,制备了均匀的纳米环。研究发现随着水的加入,聚肽均聚物首先形成纳米纤维,然后纳米纤维缠绕形成纳米环。进一步研究发现,对于聚肽均聚物纳米纤维,通过向其水溶液中加入有机溶剂的方法,得到了8字形、麻花状等新颖结构纳米环。在溶剂添加导致的成环过程中,聚肽侧基的舒展程度下降,产生了内应力驱动纳米纤维卷曲并扭转,形成了8字环等扭转的纳米环。基于以上发现,归纳了纳米纤维缠绕成环的两种典型路径:外源性作用力(如界面能变化)驱动和内源性作用力(如基团排列改变)驱动。进一步,通过聚肽均聚物与嵌段共聚物共组装,制备了一种核壳型表面螺纹纳米环,其中PBLG 均聚物构成环状内核,嵌段共聚物形成表面螺纹。采用两步组装法,即先以聚肽均聚物自组装形成纳米环,然后嵌段共聚物以纳米环为模板形成表面螺纹,得到与一步共组装法类似的结构。在聚肽纳米环可控制备的基础上,研究了其细胞内化行为。发现了包覆、成孔和胞吞三种不同的内化方式,其中,内径适中的纳米环可以镶嵌在细胞膜上(磷脂分子穿过环的内孔)从而形成稳定的纳米孔。这一特殊的纳米粒子细胞内化现象为新颖细胞膜成孔方法的发展提供了理论指导。.此外,还研究了基板介导的聚肽自组装和聚肽胶束的超分子聚合等行为,并探究了内在其机理。研究发现,聚肽共聚物在球状和平面盘状模板表面可以分别形成三维和二维阿基米德螺线等图案。聚肽胶束之间可以发生头尾相连“聚合”形成纳米线,或通过肩并肩“聚合”形成纳米束。另外,聚肽胶束可以附着在基板表面作为种子,引发溶液中的聚肽胶束发生聚合形成纳米线。
