The feasibility of using a TLN rotor (Beckman Instrument Co.) for lipoprotein separations was examined. This rotor holds tubes at a 9° angle from the vertical, a condition which reduces macromolecular binding to the sides of centrifuge tubes. A procedure for resolution of albumin, high-, low- and very low-density lipoproteins in 45 min, using the TLN rotor and 13 × 38 mm centrifuge tubes. While some overlap of fractions is unavoidable in such a small volume, appropriate corrections can be made. This procedure was used to examine the distribution of 4 drugs in human serum as a function of the total serum cholesterol content.
