The colorectal cancer (CRC) associated 40 kD glycoprotein defined by monoclonal antibodies (MAb) C017-1A (1) and GA733 (2), which bind two different epitopes on the antigen (3), is a valuable target for active immunotherapy in these tumors. The antigen is differentially expressedby gastrointestinal carcinomas as opposed to normal tissues of the gastrointestinal tract (1, 4-6). It is not shed by tumors into the patients' circulation (7, 8). Recently, passive immunotherapy of colorectal carcinoma patients with MAb C017-1A, either alone or in combination with GM-CSF, has demonstrated antitumor effects of targeting of this antigen (9, 10). In approaches to active immunotherapy of the C017-1A/GA733 antigen we have developed anti-idiotypic antibodies (Ab2) against anti-CRC MAb C017-1A and GA733 (Abl) in goats (11, 12). Both Ab2 induced in animals anti-anti-idiotypic antibodies (Ab3) that specifically bound to tumor cells and isolated C017-1A/GA733 antigen and shared idiotypes with the Abl (11, 12).
