A method for the isolation of DNA fromT. vaginaliswhich uses DEPC (diethyl pyrocarbonate) and Triton X-100 is described. DNA isolated in this way cut well with restriction enzymes and reacted with T4 DNA ligase. It is considered that this method is effective because of strong nuclease inhibition by DEPC combined with a fractionating effect of centrifugation in 1% (v/v) Triton X-100 that separates the nuclei from the bulk of the nuclease activity.
