Since publication of the first spectrum of a nucleoside more than 25 years ago (I), mass spectrometry has successfully been applied to a wide variety of problems in nucleic acid chemistry. The areas in which mass spectrometry has found greatest application are (in approximate order of literature citations):(1) the structure elucidation of nucleic acid components isolated from biological sources, especially the identification of modified nucleosides in tRNA hydrolysates, nucleoside antibiotics, and nucleosides or nucleic acid bases or both in human urine;(2) for the quantitation of nucleic acid analogs (antimetabolites) of medicinal importance in the treatment of cancer and other diseases; and,(3) most recently, the determination of the structure of complexes formed between the nucleic acids and drugs or suspected carcinogens.Information obtained in each of these three areas has had a significant impact on the knowledge of nucleic acid chemistry, ranging from a better understanding of the biochemical processes involved in nucleic acid biosynthesis to improving the clinical utility and design of antitumor agents, and other classes of medicinal agents that are analogs of naturally occurring nucleic acid components. A number of factors are responsible for the continuing, and increasingly important, role of mass spectrometry in the analysis of nucleic acid components. First, of all currently available analytical techniques, only mass spectrometry combines the sensitivity and specificity needed for the analysis of microgram (pg) and submicrogram quantities of sample with the highly informative, structurally specific information contained in the mass spectrum. The value of mass spectrometry in the structure elucidation of an unknown nucleoside is evident in the many literature citations where the total amount of material available for analysis is in the low microgram range. Likewise, the sensitivity and specificity of mass spectrometry is of primary importance in the quantitation of nanogram (ng) and subnanogram levels of potent nucleoside antitumor agents in the blood of cancer patients.
