Metabolic and functional (heart only) effects of long-term (15 months), low level (5 parts/106in drinking water) cadmium and/or lead ingestion on myocardial, hepatic and renal tissues were assessed. Myocardial peak active tension was lower and tissue heavy metal levels were elevated significantly in cadmium and/or lead-exposed rats. The ATP (P< 0.01), the total high energy phosphate compound (ATP + PCr) (P< 0.05) concentrations and the ATP/ADP ratios (P< 0.02), detected by phosphorus-31 nuclear magnetic resonance spectroscopy, in myocardial tissues from mildly hypertensive rats fed cadmium, lead, and lead + cadmium were depressed. Apparent cation-specific changes in myocardial metabolism involved glycerol 3-phosphorylcholine (cadmium; 25% of control) and dinucleotide (lead; 229% of control) metabolism. Analysis of hepatic and renal tissues demonstrated cadmium-specific or lead-specific biochemical alterations which were similar qualitatively to the changes observed in cardiac tissues. Additive rather than synergistic effects were generally observed in tissues from lead + cadmium-fed rats; however, cardiac phosphocreatine (P< 0.02) and hepatic sugar phosphate (P< 0.01) concentrations were depressed. Compounds, presently of undetermined chemical composition, were readily detected in cardiac and hepatic samples from cadmium- and lead + cadmium- but not lead- or control-fed animals.
