EW techniques for studying nucleic acids are changing our N understanding of many problems in the genetics and cell biology of ciliates. I will try to give you an update on 4 topics: serotypes, the macronucleus, caryonidal inheritance and the newly discovered technique of transformation in Paramecium. Some of the work reported here was done in my laboratory and in the laboratory of Barry Polisky at Indiana University with the help of graduate students and visitors. However, current studies are being carried out by a number of workers around the world, for example, Francois Caron and Eric Meyer at Gif, France, Jim Forney at Purdue University (formerly in the laboratory of Elizabeth Blackburn at Berkeley), Helmut Schmidt in Miinster, Germany, Karl Aufderheide at Texas A&M University, Sadaacki Kozumi in Sendai, Japan and John Sommerville in St. Andrews, Scotland.SEROTYPES Every Paramecium is covered by 1 of a family of proteins called the “immobilization antigens.” Different clones of paramecia of the same genotype often differ in their immobilization antigens and are said to be of different serotypes, each being immobilized by its own homologous antiserum, but not by antiserum prepared against the others. Eleven different serotypes, A, B, C, D, E, G, H, I, J, N and Q, have been identified in stock 5 1 of Paramecium tetraurelia. Most of the serotypes are relatively stable when cultured under standard conditions, but can be induced to switch to other types by changing cultural conditions, such as temperature, amount of nutrients, etc. All types show mutual exclusion, each Paramecium bearing only 1 antigen at a time. The antigens are large, single polypeptides varying in molecular weight from 240,000 to 320,000. The function of the immobilization antigens is unknown. Because of their very large size and abundance, the proteins and their mRNA are easily identified on gels and easily isolated [12, 281. Genetic studies [reviewed in 26, 331 have demonstrated a series of unlinked genes for immobilization antigens and the mRNA have been used as probes to isolate the genes from lambda phage genomic libraries. The genes for serotypes A, C, D and H have been the most studied genes in P. tetraurelia, while serotypes G and D are best known in P. primaurelia.
