Many metal nanoparticles are reported to have intrinsic enzyme-like activities with great promise for chemical and biomedical applications. In this work, we report PtCu alloy nanoparticles, synthesized through hydrothermal treatment of Cu2+ and Pt2+ in aqueous solutions, with ferroxidase- and peroxidase-like activities. Using electron spin resonance (ESR) spectroscopy and colorimetric method, we found that PtCu NPs exhibit strong ferroxidase-like activity in a weakly acidic environment and the activity is not affected by temperature or many other ions except silver ions. The ferroxidase-like activity of PtCu NPs was then applied for the specific detection of Fe2+ in Puxue oral liquid based on the color reaction of salicylic acid with Fe3+. In addition, PtCu NPs was found to be able to catalyze H2O2 and generate hydroxyl radicals, exhibiting peroxidase-like activity. This helps understand the underlying antibacterial mechanism of PtCu NPs against Staphylococcus aureus and Escherichia coli. Our study prove that the ferroxidase- and peroxidase-like activity of PtCu NPs are convenient and efficient for Fe2+ detection and antibacterial applications.
