Human brain vessels are known to receive a rich supply of adrenergic and cholinergic nerve fibers, demonstrated by the use of formaldehyde fluorescence and acetyl cholinesterase histochemistry 6. The rapid development of immunohistochemical methods during the last decade has allowed for studies on neuronally stored peptides 13. Thus, the first neuropeptide to be found in nerves around cerebral vessels was vasoactive intestinal polypeptide (VIP) a, 15. Potent effects of VIP have been observed on the cerebral circulation; VIP is able to relax pial arteries in vitro and in situ 15,~ 6, 22, to increase the cerebral blood flowll, t6, 23 and to enhance the cerebral metabolic rate of oxygen 6.Recent immunohistochemical studies have shown that yet another neuronal peptide substance P (SP), occurs in a well-developed network of delicate, varicosed nerve fibers in pial arteries from laboratory animals2, 4, 2L The studies have furthermore shown that SP may dilate feline pial arteries both in vitro and in situ 4. The present study was undertaken to evaluate whether SP-containing nerve fibers occurs also in human brain vessels and to investigate the pharmacological effects of SP on human pial arteries.
