Tissue imaging has the potential to be immensely useful in areas as diverse as detecting cancer boundaries and mapping signaling molecules in brain tissue. In order to retain the information on the location and assembly of the molecular makeup of tissue, the materials should stay unadulterated to the largest extent possible. The sample preparation and acquisition conditions, especially the use of solvent and vacuum, 1 may become detrimental. Here, we report the analysis of mouse brain tissue by solvent-free matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) using a field-free (FF) transmission geometry (TG) atmospheric pressure (AP) source design. Imaging by MS is well established, especially using secondary ion mass spectrometry (SIMS), but it is only marginally useful with intact biological tissue. MALDI MS, however, has been employed for tissue imaging with success, especially for high-abundance components such as membrane lipids, drug metabolites, and proteins. 2 Spatial distributions of 100 mm are routinely achieved. 2 However, there are a number of disadvantages in using vacuum-based MS for tissue imaging, especially in relation to unadulterated tissue (Fig. 1). AP MALDI tissue imaging circumvents many of the disadvantages of vacuum MALDI but has been limited because of sensitivity issues. 3 Desorption electrospray ionization (DESI), in which ions are produced by a liquid spray at AP, is currently being explored as a method for tissue imaging, but the resolution currently available (200mm) is limited by a number of factors. 4 Improvements in mass-specific tissue imaging would lead to numerous biological applications.Major challenges associated with imaging by MALDI MS include the degradation of the spatial resolution upon deposition of the MALDI matrix, interferences and suppression caused by the multitude of compounds in these complex samples including the natural abundance of salts, insufficient detection sensitivity, the limited dynamic range of a purely MS-based analysis and problems associated with placing tissue in vacuum. Quantitation is problematic and, in addition, the matrix background can interfere in the MALDI analysis of small molecules (eg drugs). In spite of the superb inherent sensitivity of MALDI MS, 5 the complexity and concentration range of various biological compounds in
