The existence of a sodium-calcium exchange mechanism was first inferred from calcium-flux measurements on guinea-pig atria by Reuter & Seitz (47). Subsequently, N a-Ca exchange received much attention due to the critical importance of Ca transport in cellular regulatory pathways. Na-Ca exchange may be essential in mediating several Ca-dependent responses, with the most prominent example being regulation of the cardiac contractile state. In addi tion, Na-Ca exchange may play a role in maintaining the low cellular Ca level in several tissues. Cells possess multiple Ca-transport mechanisms, however, and unequivocal assignment of a Na or Ca flux to Na-Ca exhange is often not possible in intact tissues. The recent development of techniques for the measurement of Na-Ca exchange in isolated plasma membrane vesicles has greatly facilitated investigations, and this review will focus solely on these studies. Na-Ca exchange has now been described in plasma membrane vesicles from several tissues, but the most detailed measurements have been made using vesicles of cardiac sarcolemma and, to a lesser extent, brain plasma membrane. Reviews describing pioneering studies of Na-Ca exchange in more intact preparations have recently been published (6, 12, 25, 46).
