The current definition of mitochondrial compartments derives from a liason established in the 1950’s between biochemistry and electron microscopy. The double membrane nature of this organelle was initially recognized by electron microscopists around 1953 (see Sjostrand 1977). A few years later, biochemists divided the mitochondrion into sucrose-penetrable and -nonpenetrable volumes (e.g., Werkheiser and Bartley 1957) which were assigned in collaborative biochemical/ultrastructural studies to the inter-membrane and matrix spaces respectively (Pfaff et al. 1968). (However, these assignments were not unambiguous; see Tedeschi 1965).
