The response of the organism to infectious pathogens involves a complex interaction between immune and inflammatory cells which are regulated by a network of cytokines. Cytokines play an important role in the interaction between effector cells and accessory regulatory cells during host defence mechanisms in response to pathogens, both in antigen-specific, adaptive immunity and in innate or natural resistance. The mechanisms of natural immunity, which possesses a low level of specificity, can be activated within minutes or hours after infection and provide a first, non-specific, line of defence. Most of the time, however, it is only with the induction of the adaptive (specific) immune response within a few days that clearance of the infectious agents, eg bacteria or viruses, is accomplished. In some circumstances (particularly following infections by extracellular parasites), the specific effector response is mainly based on the production of soluble antibodies (humoral immunity). In contrast, cell-mediated immune reactions predominate in response to intracellular parasites.The mechanisms responsible for this dichotomy have been made clearer by the observation that both murine and human CD4+ T helper (Th) cells comprise functionally heterogenous populations based on their profile of cytokine secretion (Mosmann and Coffman, 1989; Romagnani, 1991). Thl cells produce IFNy and TNFP, whereas Th2 cells produce IL4 and IL5. Other cytokines, such as IL2, IL3, GM-CSF, TNFa, IL6, IL10 and IL13, particularly in humans, are variably produced by both Th cell subsets. After the discovery that IL12, a product of activated macrophages, was a powerful inducer of IFNy production by both T lymphocytes and NK cells (Kobayashi et al., 1989), we hypothesized that IL12 released in response to macrophage infection by intracellular parasites could play a key role in the differentiation of Th cells into the Thl-like subset
