Endothelin is a newly described novel polypeptide that has been isolated from cultured porcine aortic endothelial cells (Yanagisawa et al., 1988). Endothelin produces systemic hypertension in the rat and has recently been shown to possess marked contractile activity on isolated rat renal arterial segments (Tomobe et al., 1988; Yanagisawa et al., 1988). Since this peptide had greater contractile activity on renal arterial segments from spontaneously hypertensive rats (SHR) than from Wistar-Kyoto rats (WKY), a role for endothelin in altering the renal circulation in hypertension was suggested (Tomobe et al., 1988). In the present study, the effects of intravenous (iv) and intrarenal (ia) injections of endothelin on systemic and renal vascular resistances were investigated. Seven cats of either sex (1.7-2.2 kg) were sedated with ketamine, 10 mg/kg im, and anesthetized with pentobarbital sodium (10 mg/kg iv). The trachea was intubated and a femoral vein and brachial artery were cannulated to permit drug administration and measurement of systemic arterial pressure, respectively. Cardiac output was measured by thermodilution technique, injecting 1 ml of normal saline (22-24 o C) into the fight atrium and computing cardiac output by detecting blood temperature changes with a thermistor catheter in the ascending aorta.
