InferleAn I (IL-IJ, o cyfokine zuitlz diverse actions, hns been proposed ns a mediator of both be~ zeficiul and defrimenhl responses to inflammntion and injury. Marry of tlze actions of IL-?, srrch as fhose on behadour, neuroendo-crine funcfion, sleep, fever and metabolism, are mediated by fhe CNS, as described here by Nancy Rothwell. IL-1 can be synthesized and ocf locally wifhitz tke brain to inflzrence neuronnl and giial funcfion, and has been strongly implicofed in rtormnf brairz developmenf and responses to bruin injury. A nzrnzber of disfizzcf sifes and mechanisms of acfion have beerz proposed to explnb these diverse effecfs of IL-2 in fhe brain, probably irzvolving mzrltiple receptor subtypes and complex infemcfions wifk neurotransmiffcrs~~ z (i rzeurodepfides. lnterleukin 1 (IL-l), a 17 kDa peptide, exists in two forms, IL-la and IL-la, which are separate gene products sharing only about 26% sequence homolog) r2. lL-I@ is the major secreted form, ap pearing in plasma and tissue fluids, and is the predominant molecule in brain tissue. IL-1 was one of the earliest identified members of a group of peptides now known as the cytokines, which exhibit diverse and overlapping activities on biological functions, largely related to immune function or aspects of the acute phase response (the cascade of local and systemic events immediately initiated by the body on infection or injury). However, within the past few yea-. interest in cytokine biology, and in IL-l particularly, has expanded with the demonstration of potent actions on neuronal function, behaviour, neuroendocrine systems and metabolism, all of which appear to be mediated by direct actions on the CNS.
