In addition, that of the cells treated for 3 h with 30 ng/ml of thapsigargin and then cultured in thapsigargin-free medium was also inhibited (Fig.A) . In either case, the cells became small with narrow cytoplasm and fragmented nuclei after 4 days of culture. Although more than 98% of these cells were still capable of excluding trypan blue dye, DNA fragmentation was recognizable already at this stage (Fig. B). Then, the cells gradually detached from the culture dish and were dead. Thapsigargin elevated [Ca of control cells from 118 to 461 nM. EGF, a stimulator of hepatoma cell growth, elevated [Ca2+]i of control cells to 435 nM, However, EGF did not increase [Ca2+]i of cells precultured for 3 days with 5 ng/rnl ofthapsigargin (Fig. C),
