The ADAP-SKAP55 proteins form a molecular complex that regulates adhesion and migration of T cells. Our findings from the 2nd funding period suggest a role of the two proteins in modulating membrane scaffolding and actin filament organization. We now plan to delineate their contribution to actin-mediated migration of T cells by structural, biochemical and molecular biology techniques. We will exploit our mechanistic insights to probe in mice the role of ADAP-SKAP55 as well as their interaction partners in stress-induced T cell extravasation into the brain meningeal space and clarify their relevance for T cell-supported cognitive functioning and coping with traumatic stress.
