Chloroplasts encode a small set of proteins which are however important for the biogenesis of photosynthetic complexes and the gene expression machinery. Their expression requires a variety of nuclear encoded factors. Recently, we have identified two novel proteins involved in plastid gene expression. PTAC12 is a protein of unknown function but has been discovered as component of the transcriptionally active chromosome suggesting a role in transcription or post-transcription. PPR10, a pentatricopeptide repeat protein, acts as RNA binding protein which is involved in RNA stabilization and maturation of photosynthesis genes. Using molecular and biochemical tools the project aims to elucidate the physiological function of PTAC12, to identify its native nucleic acid ligands on a genome-wide scale and to analyze its potential environmental light regulation via redox signals. A proteomic-based approach focuses on the identification of potential protein interaction partners of both PTAC12 and PPR10. New proteins will then be characterized in planta. The project aims to advance understanding of DNA/RNA metabolic processes and to achieve insights into the role of new proteins involved in plastid gene expression.
