Chlorella viruses code in their genome for a K+ channel. Data support the hypothesis that this channel is present in the virion. During infection it is inserted into the plasma membrane of the Chlorella host cell and causes a depolarization (¿Em). Selective block of the channel inhibits the virus-induced ¿Em and prevents viral replication at the step of DNA ejection. Aim of the present project is to understand the physiological importance of the viral channel generated ¿Em. Experiments are designed to examine its significance in lowering the turgor pressure of the host and the consequence of this on DNA ejection from the virus. Further experiments will address the question whether the ¿Em has a consequence on the ion milieu of the host cell cytoplasm. This could play a role in the strategy of the pathogenic viruses in shutting off host cell metabolism. Further experiments are planned to examine the importance of ¿Em in triggering a process, which prevents poly infection. This mechanism allows an individual virus to prevent infection of a cell by a second virus. Together this project will provide basic information on the general question on how viruses enter and corrupt their host.
