In project B4 we will use supramolecular ligands to modulate Protein-Protein Interactions (PPIs) of 14-3-3 proteins. These adapter proteins exhibit an extensive interactome with more than 200 partner proteins in human cells, many of which are associated with a number of human diseases. Cell biology, biophysical PPI analysis, and protein crystallography will be employed to determine the mode-of-action and potency of supramolecular ligands developed in A1, A2, A3, A5, A10 and A11 modulating biologically relevant 14-3-3 functions.
