Project P17 investigates how guanine nucleotide exchange factors (GEFs) integrate different cellular cues to properly activate their cognate Rab GTPase in the spatio-temporal coordination of intracellular traffic. The group will elucidate the molecular basis how a particular class of GEFs, LH-GEFs, establish microcompartments by (i) biochemical and structural characterization of regulatory proteins (ii), studying membrane association of LH-GEFs through lipid interactions in vitro and sub-cellular localization in vivo, and (iii) determining the atomic structure and overall architecture of LH-GEFs.
