本研究課題の目的は流通中の野菜に対する機械的ストレスの影響を，概日リズムに基づいた光制御により低減させることである。本研究では，野菜が収穫後も維持していると考えられる概日リズムに着目し，光環境制御により野菜の品質を効果的に保持できる条件を探索する。本年度は，概日リズムの解析に用いる時計遺伝子を絞り込んだ。まず基礎的な検討としてシロイヌナズナで明らかにされている時計遺伝子と相同のブロッコリーの遺伝子１８種類を選抜しプライマーを作成した。野菜を周期的な光環境下で貯蔵できるシステムを用いて，１２時間明暗周期条件を付与した生育中のブロッコリーの本葉を一定時間間隔で採取し，リアルタイム定量PCR法を用いて遺伝子の発現量を解析した。その結果，４種類の遺伝子が異なるパターンで明暗条件に伴い変動することを確認した。その後，収穫後のブロッコリー花蕾で発現解析をしたところ，収穫前の葉とは異なる傾向を示し，時間に伴う明瞭な変動がみられなかった。収穫前後または植物体の部位により遺伝子の変動傾向が違うことが考えられた。引き続き実験材料の部位の統一や，他のアブラナ科野菜での解析を進めていく予定である。また，野菜の品質を概日リズムと関係する品質成分についての測定系の検討を行い，明暗条件，時間による成分量の変化について予備実験を実施した。仮実験で得られた知見をもとに測定系，解析手法の改良と，対象作物の選定を進めている。
