胚性および癌幹細胞のマーカーとして知られるステージ特性胚性抗原（stage-specific embryonic antigen; SSEA)-3、-4、-5の膀胱癌における発現と、その臨床病理学的意義、さらには、抗がん剤や免疫療法に対する治療抵抗性の獲得における働きについて、細胞株、動物モデル、そして、膀胱癌患者の臨床検体を用いて解析する計画を立てた。今回の研究では、SSEA-4の発現を制御した膀胱癌細胞株の樹立に成功し、その安定的な培養を可能とした。そして、その安定株の遺伝的、分子生物学的特性をPCRやウエスタンブロッティングを用いて解析した。さらに、SSEA-4と同様の手法を用いて、SSEA-3およびSSEA-5についても膀胱細胞株での発現を制御する手技に関する研究を行った。膀胱癌患者の摘出した癌組織を用いた研究では、SSEA-4の発現を免疫組織学的手法で半定量化する手技を確立し、実際に数十検体でその発現と臨床病理学的特徴および抗がん剤の効果との関連を検討した。その結果、SSEA-4はhigh grade癌や筋層浸潤癌でlow grade癌や筋層非浸潤性癌よりも高発現していることが分かった。また、術前化学療法を行った患者において、SSEA-4の発現と術前治療による癌細胞の変性の関連を検討したところ、SSEA-4を高発現していた患者では、化学療法後の細胞変性や細胞死は最小限に留まり、その抗腫瘍効果が十分でない患者で高発現していたことが示唆された。一方、術前化学療法が著効した（細胞死が顕著に得られた）患者ではSSEA-4の発現を認めなかった。このように、SSEA-4の発現が抗がん剤の抗腫瘍効果の予測因子になる可能性が示唆された。なお、SSEA-3、SSEA-5については、臨床検体において免疫組織学的に解析するための手技を検討した。
